leads the field by one shot after the opening round of the Byron Nelson Championship on the USPGA Tour.
The Texan, playing in front of his home crowd, fired a first round 65 at the TPC Four Seasons Resort to leave him on -5.
Two players are on -4 after carding rounds of 66, defending champion Brett Wetterich and Denmark's Anders Hansen.
Englishman Luke Donald has made a good start to the tournament and is joined by four other players on -3, Scott Verplank, Australia's Nathan Green and John Senden and Fredrick Jacobson of Sweden.
Pre-tournament favourite Vijay Singh and world number four Phil Mickelson are four shots off the pace after opening rounds of 69.
But O'Hair, who was runner-up at the same event two years ago, is the man they all have to catch and he is in confident mood.
The 24-year-old said: "All in all I feel pretty good with my ball-striking, and it was just a really nice round of golf.
"The wind was really brutal at the beginning, so it was playing pretty tough out there."
O'Hair is currently trading at 9.4 to become the champion on Sunday, while the youngster can be backed at 2.48 to finish in the top five.
Despite not playing a practice round this week, Mickelson heads the winner market at 9.2. Donald and Singh are on offer at 11, while Wetterich can be backed at 12 to defend his title.
